摘 要 胰岛因子(Islet1,Isl1)属于LIM同源框基因家族,其氨基酸排列序列在啮齿类动物和人中存在高度保守,在结构上具有一个同源结构域和两个串联的LIM结构域。在许多器官和组织的发育过程中和出生后都对细胞起着促进增殖的作用。在胚胎的发育阶段,Isl1受到多种上游信号通路的调节,对心脏和胰腺的形成、运动神经元的分化具有重要影响,与糖尿病的产生以及多种肿瘤有关。90100
本实验通过Cre-LoxP重组酶系统在小鼠基因打靶中的应用,敲除2月龄即成年小鼠的Isl1基因,小鼠表现出糖尿病症状。通过对出生7天小鼠做基因型鉴定,鉴别出Isl1突变体,以便于后续研究。

This experiment through the Cre - LoxP recombination enzyme system in the application of gene targeting in mice, knock out 2 months namely Isl1 gene of adult mice, Genotyping of 7-day-old mice was performed to identify Isl1 mutants for subsequent study。
