摘要:反刍动物瘤胃作为消化系统的核心部分,其瘤胃内定植了种类繁多的微生物,能够使其降解植物纤文,提供可吸收的营养物质给宿主用于生长发育,一旦瘤胃内微生物菌群组成发生明显的改变,将会直接影响动物正常的生理状态,甚至引发病变或死亡。本研究通过从相关数据库获取反刍动物瘤胃微生物不同相16S rRNA基因高通量测序数据,对比分析刍动物瘤胃不同相核心微生物菌群的差异。本试验共获取瘤胃壁、瘤胃液及瘤胃内容物样品76个,QIIME分析不同相瘤胃细菌 Shannon 平均指数分别为瘤胃内容物6.65、瘤胃壁6.09、瘤胃液6.41;Observed_species平均值分别为瘤胃内容物247、瘤胃壁168、瘤胃液209,且瘤胃液显著高于瘤胃壁(P < 0.05)。PCoA分析结果显示,瘤胃三相样品间差异明显,各自聚为一簇。物种注释结果显示,17个门、22个纲、27个目、34个科、 51个属在所有样品中被鉴定,瘤胃液中主要门为拟杆菌门、厚壁菌门和变形菌门,瘤胃上皮中主要门为拟杆菌门、厚壁菌门和黏胶球形菌门,瘤胃内容物主要门为拟杆菌门、厚壁菌门和变形菌门;瘤胃液中主要属为雷沃氏菌属、乳杆菌属和纤文杆菌属,瘤胃上皮中主要属为雷沃氏菌属,瘤胃内容物主要属为雷沃氏菌属。结果证明:反刍动物瘤胃内不同相核心微生物菌群间存在较大差异。30510 毕业论文关键词:反刍动物、瘤胃、16S rRNA基因、瘤胃液相、瘤胃固相、瘤胃壁
Analysis of microbial community in different rumen phages
Abstract: in the rumen of a ruminant animal as a core part of the digestive system, the rumen colonization of many kinds of microorganisms can degrade the plant fiber, provide nutrients absorption to the host for growth and development, once the rumen microflora changed obviously, will directly affect the normal physiological state of the animal, even cause disease or death. This study through the different types of database access from the rumen of a ruminant animal microorganism in different phases of 16S rRNA gene data, used in recent years new high-throughput sequencing technology comparative analysis of different types, different growth periods and different rearing environment, significant ruminant animals of different rumen microbial flora is the core of the difference in different feeding conditions. The results showed that the total number of 76 samples, the average ruminal bacteria richness index Chao index were rumen contents (Digesta) 487 (n=43), rumen wall (Epithelial) 273 (n=10), rumen fluid (Fluid) (n=23) 381; alpha persity index Shannon index for tumor of gastric contents (Digesta) 6.65 (n=43), rumen wall (Epithelial) 6.09 (n=10), rumen fluid (Fluid) (n=23) 6.41; OUT classification operation unit respectively with rumen contents (Digesta) 247 (n=43), rumen wall (Epithelial) 168 (n=10), rumen fluid (Fluid) (n=23) 209. After species annotation, 17 phyla, 22 classes, 27 orders, 34 families, 51 genera were identified in all samples and relative percentages were determined. The results showed that there were great differences among different core microorganisms in the rumen of ruminants
