摘要:热激转录因子(HSF)普遍存在于整个生物界中,热激转录因子通过调控热激蛋白(Hsps)的表达响应热胁迫。本实验以茶树(Camellia sinensis)品种‘安吉白茶’为材料,在其转录组数据的基础上,检索和拼接获得茶树HSF家族的转录因子基因序列CsHSFA1和CsHSF-A8。采用RT-PCR方法从‘安吉白茶’中克隆获得CsHSF-A1和CsHSF-A8转录因子基因。通过序列分析,CsHSF-A1开放阅读框有1032bp编码343个氨基酸,CsHSF-A8则有837bp编码278个氨基酸。根据实时定量PCR分析表明,在‘安吉白茶’中CsHSF-A1和CsHSF-A8在高温(38 ℃)和低温(4 ℃)胁迫下的诱导表达,且表达量呈现不同的变化,CsHSF-A1只响应高温胁迫,CsHSF-A8对高低温胁迫均有响应。本研究为茶树中热激转录因子提供了总结也将为之后的进一步研究高温胁迫调控机理提供了参考。37997 毕业论文关键词:茶树;温度胁迫;HSF转录因子;表达调控
Clone and Expression on CsHSF-A1 and CsHSF-A8 in Camellia sinensis
Abstract:The heat shock transcription factors (HSFs) widely exist in the whole biology world, HSFs regulate heat stress response by regulating the expression of heat shock proteins (Hsps). The CsHSF-A1 and CsHSF-A8 gene sequences, which encoding two HSF transcription factors, were isolated based on transcriptome and genome sequence data of tea plant (Camellia sinensis). The full-length of the open reading frame of CsHSF-A1 was 1032 bp, which encoded 343 amino acids. The full-length of the open reading frame of CsHSF-A8 was 837 bp, which encoded 278 amino acids. Quantitative real-time PCR analysis of the expression profiles showed that the CsHSF-A1 and CsHSF-A8 genes could be induced by low (4 ℃)or high (38 ℃)temperature treatments. The expression levels of CsHSF-A1 and CsHSF-A8 genes exhibited difference in tea plant cultivar of ‘Anjibaicha’. CsHSF-A1 only to high stress, CsHSF-A8 to low or high treatments. This study provided a relatively detailed summary of HSFs in tea plant and may serve as a reference for studies on the mechanism of temperature stress regulation by CsHSFs.
