摘要近年来,随着影像学的发展,医学界对治疗成人股骨头坏死的研究又有了新的方法,现在应用骨髓间充质干细胞治疗成人股骨头坏死也有了新突破。骨髓间充质干细胞是存在于骨髓组织中的一类成体干细胞,来源于发育早期的中胚层和外胚层。利用骨髓间充质干细胞分化的多项潜能,来改善股骨头坏死区域的血液循环状态,促进病灶部位的血管的再生,以达到早期彻底治疗股骨头坏死的目的。骨髓间充质干细胞跨系、甚至跨胚层横向分化的可塑性及其强大的分化潜能使得它可为组织修复提供可能性。但有报道发现干细胞植入与传统髓心减压法疗效无显著差异,其原因可能在单纯干细胞的植入缺乏良好的骨支架材料。Bio-oss 骨粉是一种自体骨替代物质及骨组织再生支架,具有良好的生物相容性。也是一种很有前途的股骨头坏死的骨支架材料。实验通过原代取材、传代培养、共培养及诱导分化、microRNA提取及检测等,对Bio-oss 与骨髓间充质干细胞BMSCs共培养做相关基因分析。实验结果显示:bio-oss骨粉与细胞共培养后 miRNA-214 的表达上调。因此后续实验应用agomiRNA-214(miRNA-214 激动剂) 或antagomiRNA-214(miRNA-214 拮抗剂) 处理细胞,再与Bio-oss 骨 粉共培养,在动物模型体内试验考察 Bio-oss 骨粉联合处理后的细胞移植对骨坏死组织的修复效果。76275
