摘要  蝉花是我国的一种传统的珍贵药材,与冬虫夏草的成分和功效相近,有着悠久的食用药用历史。由于近年来冬虫夏草资源不断开采,使得价格直线上升,助推了人们对蝉花的需求,而蝉花生长需要适宜的温度、湿度、光照和昆虫寄主。近年来,蝉花的人工培育逐渐成为研究的热点,然而至今为止蝉花的规模化培育尚未取得突破性进展,主要的难题是如何能够筛选到优质的菌种资源,提高蝉花菌株对宿主的感染率。81708

本研究从采集于浙江安吉竹林的野生蝉花中分离,经过多代筛选获得能够感染蚕蛹的蝉花菌株,我们以蚕蛹为材料培育蝉花,优化蚕蛹蝉花培育工艺条件,并对蚕蛹蝉花中功能性成分进行测定,结果发现蚕蛹蝉花人工培育最适条件为22℃暗培养至蚕蛹变硬节间出现白色菌丝体,后进行22℃明暗交替培养,光照培养时湿度为95%;以家蚕蚕蛹为寄主培育蝉花,孢子悬液浓度为104/mL时,子实体长势最好,为最适浓度。以柞蚕蚕蛹为寄主培育蝉花更易形成子实体且子实体生长粗壮有力,优于家蚕蚕蛹;未形成孢子蚕蛹蝉花腺苷含量高于野生蝉花,孢子较多的蚕蛹蝉花虫草酸含量高于未形成孢子蚕蛹蝉花和野生蝉花。上述初步研究结果表明柞蚕蛹是一种理想的培育蝉花的基材,具有潜在的诱人的应用前景。

ABSTRACT Cordyceps Cicadae is a kind of precious medicinal herbs with similar composition and efficacy of Ophiocordyceps sinensis, which can be used as a substitute for O。sinensis。In recent years, Due to the shortage resources of O。sinensis, the prices rose in a straight line, boosting the people demand for C。cicadae。 But C。cicadae growth needs appropriate temperature, humidity, illumination and entomophagous parasite。 Artificial cultivation of C。cicadae has gradually become the focus of  researches。 However, so far C。cicadae scale cultivation has not been successful, the main problem is how to select high quality strain resources, and to improve the infection rate of C。cicadae to host。

In this study the strain was isolated from Zhejiang Anji bamboo, after several generations of selection, the C。cicadae strain infected silkworm was obtained。We used silkworm pupae as material to cultivate C。cicadae, optimizate artificial cultivation conditions and determinate functional components for cultivated C。cicadae。 Results showed that the optimum conditions for artificial cultivation of Cordyceps cicadae was 22℃ dark culture,until  pupa harden and internodes appear white mycelium, and then translate to 22℃ alternating light and dark culture, light culture humidity was 95%。 Silkworm pupa as host to cultivate C。cicadae, when spore suspension concentration is 104/mL, the fruiting body grows best,which is the most suitable concentration。 Relative to the silkworm pupa, tussah pupa as host to cultivate C。cicadae is easier to form fruiting bodies, and the fruiting body is stronger and better than that of silkworm pupa。 Adenosine content of cultivated silkworm pupa C。cicadae without spores is higher than that of wild C。cicadae and silkworm pupa C。cicadae with more spores; However,  the conclusion is the opposite for the cordycepic acid content。 The above research results indicate that the Tussah silkworm pupa is the ideal materials for producing Cordyceps Cicada artificially, which has potential application in the future。
