摘 要 盐酸克林霉素棕榈酸酯是新型林可霉素类抗生素,是克林霉素和棕榈酸的酯化物的水溶性盐酸盐。目前工业生产盐酸克林霉素棕榈酸酯采用传统的化学方法,而新兴的脂肪酶催化下的一步合成法成为了当下研究热门。脂肪酶因其广泛的存在而倍受研究者的青睐。现如今已广泛的应用于有机化学、食品、生物技术、制药等诸多领域。本实验通过水相与离子液体体系中用脂肪酶成功催化合成盐酸克林霉素棕榈酸酯。脂肪酶催化转酯化反应表现出的高度区域选择性,因而使盐酸克林霉素棕榈酸酯的合成方法更加广泛。同时该实验也证实离子液体能简单的提高酶的活性,本实验采用1-丁基-3-甲基咪唑六氟磷酸盐和水组成混合体系来进行酶催化反应,离子液体与水相比例为13:3。5,反应10h后测得液相色谱图在25min左右出现产物峰,产物峰的出现证实了水和离子液体的混合体系确实能催化反应。90138
Clindamycin palmitate hydrochloride is a new type of lincomycin antibiotics, clindamycin and palmitic acid ester water-soluble hydrochloride。 The industries used to produce clindamycin palmitate in a traditional way while a new lipase which can catalyze clindamycin in one step becomes the current popular research topic。 Lipase is favored by researchers due to its extensive existence, and has been widely used in organic chemistry, food, biotechnology, pharmaceutical, and many other fields。 Our experiment catalyzed the synthesis of clindamycin palmitate through the water phase in the solvent system and ionic liquid system under the lipase。 Enzyme-catalyzed transesterification reaction showed high regioselectivity, thus making the clindamycin palmitate synthesis more extensively used。 The experiment also confirmed the theory that ionic liquid can improve the activity of the enzyme。 We used 1-butyl-3-methylimidazolium hexafluorophosphate and water mixture system for enzyme catalytic reaction。 Ionic liquids and aqueous phase ratio is 13:3。5, and the reaction after 10 hours showed liquid product peak at about 25 minutes。 The appearance of product peak confirmed that the mixture of water and ionic liquid system can really catalytize reaction。
