摘要:通过查阅相关资料,我们了解到硫代膦酸脂类化合物在很多领域都有很重要的作用,探索如何优化反应条件,进行各种底物测试,对反应的普适性进行相应探讨,从而优化合成方法是本次实验的目的。81979
本论文首先选择2-(4-溴乙基)异二氢吲哚-1,3-二酮对Rp-L-薄荷氧基苯基氢亚膦酸脂、硫粉反应,进行模型反应硫代膦酸酯类化合物的合成。反应通过水作溶剂,为获得最优的反应条件,在一定条件下,筛选反应的温度、反应的时间、反应用的碱的类型和用量、反应配比、底物的类型等反应条件是否影响反应产率的高低。我们通过对反应的条件的筛选发现,在三乙胺、H2O、50 oC的条件下,以2-(4-溴乙基)异二氢吲哚-1,3-二酮为原料、和Rp-L-薄荷氧基苯基氢亚膦酸脂硫粉反应,能得到了含P-S-C键的预期目标产物——硫代膦酸酯类化合物。之后通过相关图谱检测和分析,检测得到了目标化合物。并通过对改变反应底物判断反应的普适性,从而确定合理优化的最佳反应。

Study on the Synthesis of Chiral Thiophosphonate Compounds

In this paper, we firstly try to use 2- (4-bromoethyl) isoindoline-1,3-dione, Rp-L- menthol hydrogen-phenyl phosphonite and Sulfur powder as model reaction to construct Thiophosphonate compound reaction。In this reaction, water was chosen as the solvent, and the reaction conditions were obtained by screening condition as the reaction temperature, the reaction time, the type and amount of the base, the ratio of the reaction feed, the type of substrate and so on to obtain the optimum reaction conditions。 By screening the conditions of the reaction, we found 2- (4-bromoethyl) isoindoline-1,3-dione was used as the starting material under the conditions of triethylamine, H2O and 50 degree Celsius, and Rp-L-menthol-phenyl ethylene hydrogen phosphonate reaction of sulfur powder can be obtained the desired target product thiophosphonate compound。After the analysis of the phosphorus spectrum, hydrogen spectrum and carbon spectrum, the target compound was detected。 And universal response by determining changes to the reaction substrate, the reaction to determine the optimal rational optimization。
