摘要:乳氟禾草灵是一类使用广泛的除草剂,作用速度快、活性高,能有效防除多种阔叶杂草。但乳氟禾草灵残留期较长,污染土壤和水体环境,且具有明显的蓄积毒性。本文从长期生产乳氟禾草灵的工厂的污泥样品中筛选分离到一株乳氟禾草灵降解细菌,命名为S10。通过形态学观察、生理生化特性测定以及16S rDNA基因序列系统发育分析,将S10鉴定为芽孢杆菌属(Bacillus sp.)。实验结果表明,接种量为5%时,含50 mg/L乳氟禾草灵的基础盐液体培养基接种菌株S10,3 d后的降解率为84.3%。加大接种量和添加加营养碳氮源可以促进乳氟禾草灵的降解。菌株S10降解乳氟禾草灵的最适温度为30 ℃,最适pH为7.0。26633 毕业论文关键词:乳氟禾草灵;生物降解;芽孢杆菌属S10
Isolation, Identification and Characteristics of Lactofen-degrading Bacterium S10
Abstract:Lactofen is a kind of widely used herbicide, could effectively control a variety of broadleaf weeds. But the pesticide residues contaminated soil and water environment, and had a significant accumulation of toxicity. A strain of bacterium designated as S10, capable of degrading lactofen, was isolated from contaminated soils by lactofen. Based on analysis of its morphology, physiological and biochemical characteristics and the result of the 16S rDNA sequence homologue, strain S10 was identified as Bacillus sp. . The results showed that this strain could degrade 84.3% of 50 mg/L lactofen in liquid medium within 3 days when the inoculation amount is 5%. The amount of inoculation and additional nutrient nitrogen and carbon source could promote the degradation of lactofen and the best conditions for phenol degradation temperature were as follows: temperature 30 °C, pH 7.0.
