摘要氨基葡萄糖又被称为氨基葡糖,是氨基取代葡萄糖的一个羟基后产生的化合 物,在健康保健、医药和食品领域都具有广泛的应用,是以市场前景广阔。正因如 此,有越来越多的学者都投入到氨基葡萄糖的研究当中来。利用发酵法生产氨基葡 萄糖的技术已经有了不错的进步,其优势也比较明显,已经有学者利用大肠杆菌作 为出发菌株进行发酵实验生产氨基葡萄糖。

本实验以欧文氏杆菌为发酵生产菌株,首先研究了不同因素:葡萄糖浓度、酵 母粉浓度、pH 以及培养时间对欧文氏杆菌发酵生产氨基葡萄糖的影响,在进行预实 验的基础上采用四因素三水平的正交实验方法,以得到发酵生产的最优条件。实验 结果表明,当葡萄糖浓度为 48 g/L、酵母粉浓度为 20 g/L、pH 为 6。5 以及培养时间为 20 h 时,氨基葡萄糖的产量最优,可达到约 0。8 mg/ml,可作为提取步骤的优先条 件。且根据 R 值的大小看出,培养时间对氨基葡萄糖产量的影响最为重要。此外,主要研究了以欧文氏杆菌的发酵液为原料,将发酵液进行预处理后,利 用离子交换原理将发酵液中的氨基葡萄糖分离提取出来。提取步骤以苯乙烯强酸性 阳离子交换树脂 001×7 装柱,先利用其与氨基的作用将氨基葡萄糖挂柱,再利用洗 脱液将氨基葡萄糖洗脱下来。经过对洗脱液浓度、进样流速以及洗脱流速的实验, 得出比较优化的提取条件::最优洗脱液浓度为 0。7 mol/L 的硫酸铵溶液,洗脱率可 达到 76。9%。最优进样流速及洗脱流速均为 0。96 BV/h,洗脱率可分别达到 96。3%和 95。8%。80976

Abstract Glucosamine, also called amino glucose, is compound that one amino replaces one hydroxyl of glucose。 Glucosamine finds a wide-range of application in health care, pharmaceutical and food industries。 For this reason, more and more researchers devote into the research of glucosamine。 The technology of using fermentation to produce glucosamine has been made good progress, its advantage is as plain as the nose on your  face。 Escherichia coli was used in fermentation experiment for the production of glucosamine。

In this paper, Erwinia tasmaniensis sp。 was used for producing glucosamine。 At first, researchs on different factors which can influence the production of glucosamine。 On the basis of preliminary experiments, four factors three levels orthogonal experiments were used to get the optimal conditions of fermentation。 Results showed that when the concentration of glucose is 48 g/L, yeast powder concentration is 20 g/L, pH is 6。5 and incubation time is 20 h, the output of glucosamine optimal reached to 0。8 mg/ml。 Steps can be extracted as a priority。 According to the size of the R value, incubation time is the most important influence on yield of glucosamine。

In addition, Ion exchange chromatography was used to extract glucosamine from the fermented broth of restructured Erwinia tasmaniensis sp。 as raw materials。 The effects of extraction conditions on glucosamine extraction were studied by static test, and cation exchange resin 001×7 was selected。 After several experiments on eluent concentration, injection velocity and elution velocity experiment, more optimized extraction conditions are obtained。 The optimal concentration of eluent is 0。7 mol/L ammonium sulfate solution, elution rate can reach 76。9%。 The optimal injection velocity and elution velocity are 0。96 BV/h, elution rate can reach 96。3% and  96。3% respectively。
