摘要为研究草鱼肠道益生菌,探索草鱼肠道菌落的构成,从草鱼肠道中筛选一株菌种,暂时编号为L,通过革兰氏染色、生化试验以及药敏试验和DNA分子鉴定等方法进行细菌形态观察、理化特征、药敏及16S rDNA序列分析等研究。结果表明L菌为革兰氏阴性菌,进一步采取PCR扩增16S rDNA基因,测序大小为 1531 bp,与不动杆菌(Acinetobacter xiamenensis)的序列同源性达 97.1%;药敏试验结果显示L菌对庆大霉素、阿莫西林、卡苄西林、头孢噻肟、头孢哌酮等药物敏感,对头孢孟多、头孢噻吩、先锋霉素Ⅳ、红霉素、阿奇霉素、万古霉素、克林霉素、替考拉宁等药物抗性较强;生化试验结果为L菌为葡萄糖氧化性,阿拉伯糖、甘露糖试验呈阳性、精氨酸双水解、赖氨酸脱羧酶、鸟氨酸脱羧酶、葡萄糖酸盐、肌醇、麦芽糖、木糖等试验呈阴性。该菌株的分离与鉴定为草鱼肠道菌落的构成研究提供了一份贡献。71685

Isolation and Identification of Acinetobacter in Grass Carp
Abstract To study probiotics grass carp, grass carp explore colonies constitute screened from a species of grass carp intestine, temporarily numbered L, by Gram staining, biochemical tests and susceptibility testing and identification of DNA molecules The method for bacterial morphology, physicochemical characteristics, sensitivity and 16S rDNA sequence analysis. The results showed that L is a Gram-negative bacteria, to take further PCR amplification of 16S rDNA gene sequencing size of 1531bp, and Acinetobacter (Acinetobacter xiamenensis) sequence homology of 97.1%; sensitivity test showed that bacteria celebration L drug-sensitive large, amoxicillin, ampicillin card, cefotaxime, cefoperazone and so on cefamandole, cefoxitin, cephalosporin ⅳ, erythromycin, azithromycin, vancomycin, clindamycin teicoplanin drugs such strong resistance; biochemical test results for bacteria L glucose oxidation, arabinose, mannose test was positive, double-arginine hydrolysis, lysine decarboxylase, ornithine decarboxylase, gluconate, inositol, maltose, xylose test was negative.
