摘要:本研究以马铃薯栽培品种Desiree的转基因株系为材料进行glgB基因表达研究。对转基因马铃薯植株进行PCR检测,结果发现glgB基因已经整合到马铃薯基因组中。转基因马铃薯植株的RT-PCR分析,表明glgB基因可以在马铃薯块茎中正常表达。对转基因马铃薯淀粉分支程度和淀粉含量测定,结果发现,glgB基因在马铃薯块茎中的表达能使转基因淀粉的分支化程度增加35%,而对马铃薯块茎中淀粉的合成没有影响。转glgB基因对淀粉颗粒的形态和大小没有明显影响。转基因淀粉的理化性质分析表明,随着淀粉分支程度的增加,转基因淀粉的峰值黏度减小,糊化温度降低,析水率降低,冻融稳定性提高。78919

Abstract: The transgenic plants of potato cultivars Desiree were used as the materials in the expression of glgB genes study。 The results of PCR detection of the transgenic potato showed that the glgB gene had been integrated into the potato genome。 Then the transgenic potato plants were analyzed by RT-PCR, and the results show that glgB genes could expressed in the tubers of potato。 The determination of the starch branching degree of transgenic potato and starch content, the results showed that the expression of glgB genes in the tubers of potato results in the starch branching degree had increased up to 35%, but the expression of glgB gene in the tubers of potato had no effect on potato tuber starch synthesis。 The glgB genes expression had no effect on the morphology and size of starch granules。 The analysis of the physicochemical properties of transgenic starch showed that with the increase of starch branching degree, the transgenic starch the Peak viscosity, the Pasting temperature and the separated water ratio was reduced, the stability of freezing and thawing was improved。
