摘要:RNAi具有特异、稳定、高效以及不改变基因组的遗传组成等特性,成为基因功能研究的重要手段。传统的RNAi载体需要经过多次酶切和多片段的定向插入,步骤繁琐,成功率不高。Gateway技术不使用限制性内切酶和连接酶,克服了传统载体构建过程中需进行的多次酶切和连接反应的繁琐操作步骤及酶切位点的限制等缺点。本研究应用Gateway技术改造双子叶植物的RNA干涉载体,经酶切验证构建的载体与预期的完全一致。39415 毕业论文关键词:双子叶植物;RNA干涉;Gateway;
Optimization of Dicotyledonous Plants RNAi Vector by Gateway Technology
Abstract: RNAi has become an important means of gene functional studies for it regulate inpidual gene express level with specific, stable and efficient manners. The traditional RNA interference vector needs several enzyme digestion and directional fragment insertion with complicated steps, The success rate is not high. Gateway technology needn’t restriction enzymes digestion and ligases to overcome cumbersome vector construction process. In this study, we optimized dicotyledonous plants RNAi vector by Gateway technology. Exactly verified constructed vector was generated as confirmed by digest test.
